Development of SSR Markers for Molecular Identification of Amorphophallus Species

Abstract: Genus Amorphophallus are perennial herbs that belong to the Araceae family. For the high glucomannan content in the underground corm, several species in this genus have been planted in many Asian countries, such as China and Japan. Although there is a long history of cultivation in China, the molecular identification method of Amorphophallus species is still not available, which hinders the plantation industry. Simple sequence repeat (SSR) has been considered as one of the powerful and informative molecular markers in identifying species and crop varieties. In this study, SSR markers of Amorphophallus species were developed by using restriction site-associated DNA sequencing, and 12 screened polymorphic markers were used for identifying four Amorphophallus species. The results showed that the allele number of 12 SSR markers ranged from two to four. These SSRs could distinguish A. yunnanensis and A. tonkinensis from the rest two species. This study gives hints to the molecular identification of Amorphophallus resources in the future.
