FTIR结合曲线拟合对万寿菊植株中酯类的研究

摘要: 叶黄素酯是以棕榈酸二酯、肉豆蔻酸二酯等为主要成分的一种酯类物质,在万寿菊中含量最为丰富.为了研究叶黄素酯在万寿菊植株中的分布情况,使用傅里叶变换红外光谱仪分别测试了万寿菊花瓣、茎秆和叶片的光谱.它们的红外光谱中均在1735 cm-1附近出现酯类物质羰基的特征吸收峰,表明花瓣、茎秆和叶片中均含有一定量的酯类物质.由于羰基吸收峰受到蛋白质吸收带的叠加影响,因此未能准确测定它的强度.为了获取羰基吸收峰的信息,曲线拟合分析技术被用于分离吸收带的叠加峰.曲线拟合分析结果显示,万寿菊植株中叶黄素酯含量由高到低的顺序是花瓣、茎秆、叶片.研究表明,傅里叶变换红外光谱可对万寿菊植株中酯类物质的含量进行快速定性分析,为万寿菊植株中酯类物质的合成和运输研究提供一种便捷方法.

Abstract: Lutein ester is mainly composed of dipalmitate and dimyristate, which is the most abundant in Tagetes erecta L.. In order to study the distribution of lutein esters in Tagetes erecta L., the Fourier transform infrared spectrometer was used to obtain the spectrum of its petal, stalk and leaf. In the Fourier transform infrared spectra of the petal, stalk and leaf, the characteristic absorption peak of the carbonyl was observed at near 1735 cm-1, which indicated that the petal, stalk and leaf contained a certain amount of esters. Because of the absorption peak of carbonyl was overlapped by the absorption band of protein, its strength could not be determined accurately. For the purpose of obtaining information on the absorption peak of carbonyl, curve fitting analysis technique was used to separate the overlapped peaks of the absorption band. The results of curve fitting analysis showed that the order of lutein ester content in Tagetes erecta L. from high to low was the petal, stalk and leaf. The results showed that Fourier transform infrared spectrum could be used to analysis the content of esters in Tagetes erecta L. quickly and qualitatively, which provides a convenient method for studying the synthesis and transportation of esters in Tagetes erecta L..
